I have a newly synthesized phosphatase inhibitor that is soluble in DMSO, but as I begin to add saline in order to dilute the DMSO a 5% concentration, the drug begins to precipitate. I have tried heating, sonicating, and adding saline 100 ul at a time while vortexing (until I reach the desired 950 ul of saline + 50 ul of DMSO it was originally dissolved in). Does anyone have a recommendation as to what may be going wrong? Today I plan to attempt to heat the saline before adding it. Also, I am adding 10 mg of the drug (crystal form) to the DMSO.
I'm not sure you are doing something wrong, it may be an intrinsic property of the compound, low solubility in water. As soon as you add it (dissolved in DMSO) to water the solvent disperses in water leaving the local concentration high and it chrystalise. You may be more lucky adding it to a big volume while mixing, as to prevent high local concentrations. Don't start with little water. It may be best to go directly to a lower concentration, eg the highest one you actually need. I can't imagine that you need it at 10g/l.
Indeed adding it to hot water may help a bit, tell us how you are getting on.
