I'm working on an optical setup to tweez cells. while setting the components, came across one ambiguity in applying suitable objective's working distance since I might use cell chamber,dish,plate or... as a cell container.
Could you possibly tell me what working distance you usually apply ?
I guess nearly any trapping experiment will be using a coverslip (or coverslip-thickness substrate) between the objective and sample. Most objectives will be expecting this coverslip and will give you serious spherical aberrations if your substrate isn't the correct thickness (high end objectives even have a correction collar to dial in the exact coverslip thickness). Therefore, your question seems to me really about limiting the kinds of substrates you'll be using to hold the cells (i.e. don't put them in a standard culture dish with >1mm bottom thickness).
There are "long working distance" objectives that could possibly get you around this limitation, but they also have a substantially higher cost and typically cannot reach the high NA you may need for your trapping experiments.
