I have extracted water based extract of a plant , the water extract was soluble in ethanol and was loaded in silica based column chromatography and elutes were taken. I would like to know what mobile phase can used to check the elutes and what spraying reagents can be used to check the visibility of the bands.
i have tried used the mobile phase chloroform : methanol (8:2) and toluene : ethyl acetate (9:1) for viewing the elutes , i have tried spraying anisaldehyde sulphuric acid as spraying agent. kindly say your suggestions.

The mobile phase and spraying agent will depend on the class of compound you are looking for - they are quite specific - you may look for previous reports on the constituents of the plant for guidance.

You can use isocratic solvent system of 15:8:4:1 i.e. chloroform:ethyl acetate: methanol:water..or 12:8:8:2 same order.You can also go for reverse phase column chromatography.
It's better to use BAW (butanol: acetic acid: water) in a ratio of 4:1:5 for TLC. It will work. 5% methanolic sulphuric acid can be taken as spraying reagent.
