TPTZ solution, 10 mmol/L in 40 mmol/L HCl: 156 mg 2,4,6 tripyridyl‐S‐triazine (TPTZ)is dissolved in 50 mL of 40 mmol/L hydrochloric acid (HCl). The reagent is stored at 4 °C and can be used for at least 2 weeks after preparation. This reagent should appear clear and colorless.
Working FRAP reagent: as needed and just before use, mix 10 mL acetate buffer with 1 mL TPTZ solution and 1 mL ferric chloride solution (scale up if larger volume is needed) and use within 3 hours. Working FRAP reagent should be pale yellow‐orange in color. Discard the working reagent if an obvious blue color is observed as this indicates contamination of reagents or equipment with free Fe2+.
