I'm looking for the isolation of polymethoxyflavones from my crude extract.
Which TLC plate is better, normal or reverse phase? And which kind of solvents giving a best separation?
Anyone worked already in this kind of polyphenols!
This link leads to a summary of different polymethoxyflavones. Although written from the viewpoint of flash chromatography, the same solvents and stationary phases are used for both column and thin layer chromatography.

You will see that flavones with a higher degree of methylation are less polar and use silica with hexane/ethyl acetate. As the degree of methylation decreases, the flavones are more polar and use more polar solvents on silica such as dichloromethane and methanol. So the answer is it depends on the degree of methylation.

Hi Dr Ayoub Najem . I think it is preferred to start from nonpolar-polar solvent with the combine of them. th general solvent usually used are hexane, ethyl acetate, chlroform, dicholoromethane and methanol.
