We are going to do a seed germination experiment but found that most of the previous research used distilled water as a control solution for seed germination. As distilled water is devoid of any nutrients then plasmolysis can occur during seed germination. Can anyone suggest what should be the perfect composition for the seed germination solution as an experimental control?

In general, biotechnical tests present rapid methods for evaluating the viability of seed samples and specifically to assess non-germinable seeds that seem to be dormant through the measurement of weak or dead tissue. The Tetrazolium test is the most common of these tests. Generally soaking treatment of seeds for 18 h at 30°C in a solution of 0/5% Tetrazolium test is a suitable treatment.

Distilled water on paper, but you should have attention on stable physic environment for all treatments
I have also found that distilled water on paper kitchen towels work just fine. The paper towels should be cut to the shape of containers used (e.g petri dishes)
for Prunus species (almond, peach, almondxpeach), we use distilled water for both methods of studying the germinative capacity of the stone. Either for soaking followed by stratification in perlite or sand or sawdust as well as for stratification in a petri dish with whatman's filter paper previously sterilized in an autoclave.

It is really dependent on the type of medium or experiments that are being used. For example; Hougland solution without excessive chemical for hydroponic condition, MS salt with low levels of sugars (0.1-0.5% suc) for agar plate experiments, tap or distilled water for soil experiments. But if you are interested to calculate germination rate, it is better to provide a medium with appropriate levels of minerals to avoid any stress on plants.

I'm using seedlings as a sample grown in Petri dish with filter paper in distilled water for secondary metabolites analysis, I want to know for many days we can consider seedlings are healthy with distilled water, without supplying any nutrient. we keep seeds for germination, after two days seedlings growth appear, within 10-12 days seedlings length touch upper cover of Petri dish.
