I hope to use ethanol in sample preparation. I want to know is it need to get same solution as the mobile phase in using HPLC. Is it not, what is suitable solution for the making standard solution and for sample preparation ?
Fucoxanthin solubility: High concentrations of Ethanol should be used to dissolve the sample in (e.g. 95/5 Ethanol/Water) as the compound's water solubility is very low. Analysis by HPLC is straightforward for the pure compound. Use of a conventional C18 phase with a mobile phase gradient high in ethanol (vs water) would provide a good starting place. Please contact an experienced and local chromatographer for assistance. The sample should be dissolved mostly in ethanol with some water (e.g. 95/5), then filtered before injection.
Praveen Rathnayaka: Learning how to research a question on your own is one of the most important skills you can learn as a student. Before inquiring online, please try a keyword search on the web (i.e. GOOGLE) to find related info. Hundreds of relevant articles, papers and analysis methods may be found in this way.
I would use whatever your initial chromatography conditions are. If your sample is not soluble in that, when injecting it into your system, it can precipitate at the top of your column. I have ruined many columns in my life in that fashion.
The solution used to dissolve the fucoxanthin standard to make fucoxanthin standard solution is not necessarily the same as the mobile phase, the solution used to dissolve the fucoxanthin standard can be a different solvent system, such as methanol, which is used to prepare standard solutions for analysis
