I am testing effect of plant extract on human cell line (hacat) infected with bacteria. My extract is prepared in methanol (10 g of plant in 100ml of methanol). What concentration of extract should I use so it may not be toxic to the cell-line. Should I infect the cells prior to treatment with plant extract or extract and bacteria can be added at the same time? I

First extract and bacteria can be added at the same time, If the extract is found to have an activity you can subsequently infect cells and then treat with the extract. For doses no precise doses to be used as the effect of the extract depends primarily of the plant.

For doses, we usually have to test different doses to find out the minimum dose that shows effect.
