Can any one help me regarding lncRNA selection and comparing them in different ages and different regions from the same tissue the best selection will depend only fold differences in different ages or we must include the probability of T-test. or any other selection criteria please, guide regarding the ideal criteria for selection and considering lncRNA.
I'm not sure I've understood your question, but if you're talking about transcriptome wide analysis, a statistical test is mandatory in order to pick good differentially expressed candidates. Beside that, other factors should be taken into account, like lncRNA basal expression levels and, if available, lncRNA biological significance for the tissue under analysis.
As always, the right method depends on te is context and should be chosen according to it.
we usually consider the basal expression level, fold change, type of lncRNA, associated proteins and test it using 3 biological repeats. i dont know if 3 biological repeats are enough or not
Hi Amr, the number of needed biological replicates depends on the variance shown by the lncRNA expression between different samples, but usually 3 reps are acceptable in qPCR.
