I want to make coupling reaction between a compound with carboxylic group and the aminethat have tetrazine.
I used HATU and DIPEA for the coupling for overnight. I get three spots may be the tetrazine degradated. So, what is the perfect base and time to conserve the tetrazine from breaking down.
Can you convert acid to Acid chloride? If you can then Schotten-Baumann Reaction would be much milder and better condition to make your amide. NaHCO3 should not decompose your amine. (conditions: RCOCl+RNH2-satd. NaHCO3 solution and EtOAc rt, 30 min). Hope that helps.
Yeah, I agree with the answer given by Dr. Mustafa Guzel. After seeing the Question, Answer in my mind is already given by him. Try to prepare the acid chloride using Excess moles of Thionyl chloride and using a base in reflux condition (If your compound is stable). Isolate Clean acid chloride by distillation method and procees as above. Good Luck.
