Am doing research on effects of elevated temperature and photoperiod on accumulation of starch in several genotypes of upland rice. I intend to pick leaf samples and check for the changes in sugar levels and enzyme activity esp. Identifying which enzymes are involved in the process of rice growth and development and ton the other extent check the photosynthetic rate under field and green house conditions .
A simple qualitative way would be to use microscopy, staining with KI/I2 solution.

I use a easy and reproducible assay using the "Total Starch Assay Kit (AA/AMG) for 100 assays by Megazyme Catalog No K-TSTA.
Megazyme web description: "For the measurement and analysis of total starch in cereal flours and food products. This kit now contains an improved α-amylase that allows the amylase incubations to be performed at pH 5.0 (as well as pH 7.0)".
