I am trying to detect some esterase activity from insects. This is a quite old method, which uses naphthol acetate in combination with a diazoblue salt. I purchased Fast Blue RR however I cannot seem to get the assay to work. I suspect the issue is the solubility of the salt, as I seemingly cannot dissolve Fast blue RR. Old publications mention the DBLS solution, however after some digging I found out it actually uses Diazoblue B salt coupled with SDS. I tried to make 1% solution of FastBlue RR in water but it does not dissolve at all. I also tried dissolving it in 0.1% Triton X phosphate buffer with the same effect. Does anyone know which diazoblue compound I should use and what is/if there is any difference between Diazoblue B, Fast Blue B, Fast blue RR and Fast blue RR salt?
