I am working in telomere biology. We follow De Lange's protocol for Telomeric FISH. But unfortunately, the probe that is PNA does not bind with the telomeric repeats, though the metaphase chromosome spreading is accurate. I want to know how to overcome this. I think the problem is in the PNA probe master mix which is about 1 year old.
