Do liquid liquid seperation followed with methanol then add ammonia till d ph changes then add chloroform again do liquid liquid separation chloroform part will contain alkaloids..... For different grades of alkaloids you have to take different solvents as well as methods.So,first tell which alkaoid you want to extract.What methodyouwant to adopt.TLC,or GLC or any other method?
if you have crude extract and need to isolate alkaloid only ,you should acidify the crude extract with dil H2so4 and then purify with chloroform and reject it ,then alkaline with ammonia solution and extract again with successive portions of chloroform to accelerate the extraction ,finally the collected chloroform extract should be distilled out and the residue is alkaloid pure . i agree also with Aparna Srivastava
Liquid-Liquid, or Supercritical might work well. Question..Is there a economic way to isolate a several alkaloids during the extraction. I'm currently using buffers and PKa values.
The simplest method is to extract plant with MeOH and spin it at 5000 rpm for 15 min and the resulting supernatant can be treated with Hexane to remove plant pigments and lipids. The aqueous phase contains alkaloids, which can be quantified colorimetrically or qualitatively using HPLC adsorption chromatography
Using water and methanol in the ratio of 6:4 has show good result for my alkaloid extraction. i used six solvents(n-hexane, chloroform,ethylacetate, ethanol, methanol and water ) to elute and after testing the fractions for the presence of alkaloid only ehtanol, methanol and water contain alkaloid. I did TLC and discovered that water methanol ration given above gave better result.
