Please add compenents( Peptone 5g, Beef extract 3g, and Glycerol 70ml) to distilled deionized water and bring volum to 1L and mix them thoroughly. Distribute 500ml of the broth into screw-capped tubes in 5 ml volum. Autoclave the tubes and the flask with the remaining broth for 15 min at 15 psi pressure 121 C° cool to 25 C°.
