I am trying to extract DNA from several epidermal cells of lima bean cotyledons following gene introduction. I've obtained about 5mg tissue and am wondering what DNA extraction kit would work best for small samples.
At first i think the best way to extract DNA (if you have time) is almost always a traditional extraction (such as using DNAzol, trizol or GuSCN+EDTA+Triton).
I work with insects which, as plants, have very resistent cells due to the cuticle. I advise you not to use a generic kit. We tried specific Qiagen kit and they're not very good, they give a low yield and impure DNA.
Promega wizard kit is not bad, but it is quite expensive...
I suggest you to use Thermo-Fisher plant kits (https://www.thermofisher.com/it/en/home/life-science/dna-rna-purification-analysis/dna-extraction/genomic-dna-extraction/plant-dna-extraction.html) or zymo research kits (http://www.zymoresearch.com/dna/microbial-environmental-dna-isolation-1/soil-fecal-plant-dna/quick-dna-plant-seed-kits).
Zymo has very good products, we use their kit to extract DNA and RNA from insect (they also have a kit to extract DNA and RNA together) and they are the most efficient amongst all the producers we tried.
