A type 2 diabetic rat model was developed with High fat diet + low dose Streptozotocin. High fat diet included a 2:3 mixture of coconut oil & partially hydrogenated vegetable oil (10ml/ kg body weight per oral) IN ADDITION to normal pellet diet(56% carbohydrates; 18.02% protein; 3.95% fat; 7.85% ash, 8.75% moisture, 5.45% fibre, 1.6% Ca and 0·85% P, w/w)
How much % of total energy of diet is given by the fat in the HFD?
Do I understand correctly that that you gave rats two diets at the same time one was coconut oil (2) and vegetable oil (3) and other diet was chow??
Did all the rats eat 100% of the fat diet and then ate the rest of their calories from chow?
The % of total energy from the high fat diet depends upon how much or each diet they consumed... I cannot calculate the energy without knowing the relative consumption. The chow only provides 4% fat but the high fat diet is 100% fat and energy density of all fats is 9 kcal/gram so you need to calculate what is eaten in calories from the chow and calories from the high fat mixture diet and then express that fat intake as a % of total calories...
If you give me the numbers I can calculate this %.

I understood that you gave fat by orogastric gavage plus chow.
It would be more appropriate to report the amount of fat ingested by rats (g/day/rat and kcal/day/rat) than the fat % in the diet. If the fat was solely offered on a diet it would be easy; you calculate the amount of fat/g of chow. However, fat was also given orally, and the amount of oil varies since it depends on rat body weight (BW).
Energy
1g fat = 9 kcal
1 g carbohydrate = 4 kcal
1 g protein = 4 kcal
2:3 coconut oil & partially hydrogenated vegetable oil
You need to know oil density. In a quick search, I found coconut oil density = 0.9259 g/ml. Let's say that both oils have the same density:
- Your dose is 10 mL/Kg BW.
- 10 mL oil = 9.259 g fat
- 9.259 g x 9 kcal = 83.331 kcal fat per kg BW
Remember that the oil dose is 83.331 kcal/kg BW, but each rat received a different amount of oil volume.
Pellet diet
Let's say you have 100 g of chow:
- 56% carbohydrates = 56 g x 4 kcal = 224 kcal
- 18.02% protein = 18.02 x 4 kcal = 72.08 kcal
- 3.95% fat = 3.95 x 9 kcal = 35.55 kcal
- 7.85% ash = 0 kcal
- 8.75% moisture = 0 kcal
- 5.45% fibre = 0 kcal
- 1.6% Ca and 0·85% P, w/w = 0 kcal
Diet calories
- 224 + 72.08 + 35.55 = 331.63 kcal/100 g (3.32 kcal/g)
Calories from fat (%)
- = kcal fat/kcal (CHO+protein+fat)
- = 35.55/(224+72.08+35.55)
- = 0.1072 (ie, 10.72% fat)
Your diet has 10.72% calories from fat.
Or you can say it has 0.3555 kcal fat/g (= 35.55 kcal/100g).
Fat ingested/rat
You have to calculate the amount of oil (mL) and chow (g) ingested in one day, multiply it by fat energy, and then sum these calories to know the energy intake/rat from fat.

Caroline Fernandes-Santos provided an excellent method that you can use to calculate % energies from dietary components.. you can plug in your consumption numbers and calculate the energy % values. Memorize Caroline's formula and use for all aspects or dietary energy calculation.
