HPLC grade water is water wherein a specific conductivity (16-18) is maintained by ELGA water machine and filtered through 0.22 micron filter to avoid the particulate matter.
HPLC is acronym for high performance (or high pressure) liquid chromatography. And HPLC-grade is definition of water purity related to minimal background or signal interference for this technique. You can check any supplier for details about purity of this grade of water.
If possible, add tags analytical chemistry and (liquid) chromatography (or HPLC) to your question to obtain more professional answer.
To minimise background or signal interference for chromatographic techniques companies provides solvents for sophisticated instruments like HPLC, GC, LC-MS etc. HPLC grade water is one of it.
It's ultra pure water with low UV absorbance, often filtered through a 0.2(2)-micrometre filter and packaged in solvent-rinsed amber glass bottles and sealed under an inert atmosphere (often nitrogen). Why? To minimize contaminants (e.g., ions, particles or bacteria) that could pollute or destroy your pump/injector and affect separation quality.
This is water at 18 W purity and QC tested by HPLC analysis. It has advantages that it is of known purity and not prepared with equipment and glassware coming from the laboratory. This makes it an ideal for making oligo dilution and buffers for PCR reactions when preventing contamination is essential.
As indicated by others the main feature of HPLC-grade water is low UV absorbance which could interfere with analyte determination. Of course this presumes you are using a UV detector !!
all of answers are good and there are interested people, Just adding that can be detect you compound in water by HPLC spectrum means no peak for water compare if you using the any other solvent (will see peak of that solvent in spectrum of HPLC)
as you know particles may damage the pump and injector. they also could plug the column. also organic contamination of water may affect the chromatograms in different ways. organic molecules may accumulate on the surface of column and they cause to decrease the access of sample molecules and solvents to the binding sites. so for all of these reasons its critical to use water with HPLC grade for your work.
p.s. HPLC grade water in persion means: Ab e ba grade HPLC
HPLC grade water is water wherein a specific conductivity (16-18) is maintained by ELGA water machine and filtered through 0.22 micron filter to avoid the particulate matter.
Various vendors have bottled water that is marked "HPLC grade". It generally means there is little UV-active impurities and dissolved ions that may interfere with HPLC-UV measurement.
Salem: No. Different water grades exist to address the needs and requirements found in different applications. For PCR analysis a special grade of water often known as: "Water, PCR Grade" should be used. "PCR grade water" is certified to be free of nucleic acid contamination (very important in PCR analysis to reduce false positive results). HPLC grade water is not processed or tested to meet that criteria.
Fatima: Effects: Ghost peaks, poor baseline quality, absorbance from non-sample material, higher UV absorbance which reduces the dynamic range of any UV detector. Contamination of the HPLC column.
