I have been centrifuging the microglia at 300g for 10 minutes at 2-4 degrees Celsius, but I feel as this may be a bit too rough on the microglia. The microglia are isolated from an adult mouse (8 weeks old). Does anyone else centrifuge adult microglia, and if so, what speed do you use and for how long?

When I was looking into cell seperation numerous papers suggested 800g with no break. I found there was fairly high mortality with this though.

I found that a speed of 300g for 5 mins was enough, but some cells were still dying at this.
