Hello . We are making external lab analysis for determination some parameters . One of them is BOD5 . In the method it's written that we need to dilute solution with seeded dilution water . I read about that it must contain micro-elements , active biomass etc ... Does anyone have experience in such kind of analyse .

Its vary with your samples. If you do analysis for waste water need dilution of samples. If water samples content low organic matter content (drinking water-well water) then no need to dilute samples just get the initial DO concentration and keep incubation but initial DO should be more than 6 ppm and if you have less DO in initially then need aeration to increase the DO in water. However, when you analyzing waste water which containing high amount of organic matter content and 5 days not enough to get accurate results. Therefore, required sample dilution with dilution solution which is containing nutrients important to microbial growth. Dilution may directly influence to the sample as diluting the amount of organic matter degrading bacteria. Therefore, if you diluting your samples or samples not containing microbes necessary to add seeds which are commercially available and helping to degrade organic matter in samples. Hope this will helpful for your sample analysis.
