Try using much more diluted sugar solution.

used lower concentrations too, but it is still not working

change the mobile phase or use size exclusion chromatography 1st, then do ur tlc

yes sir...it is a mixture  of sugars dissoved in  distill water.. and the sugars are ribose, manose, glucose, galactose,xylose

and the solvents used are n-butano,distill water,pyridine and toluene 10:6:6:1

Try the two following solvent systems :

System I (rapid migration) : n-butanol/ethanol/water/acetic acid/pyridine (10/100/30/1/10; vol/vol)

System II (slow migration) : n-butanol/acetic acid/water (2/1/1; vol/vol)

spot your sample 2 cm from the bottom of your plate, dry and then put it in the development chamber with only 1 cm height of the solvent and wait until the complete development of the plate

Dear Colleague, it will be better if you use the silica plates instead of cellulose plates with system II describe by the Dr Karamanos just before my answer

Our best results are with silica gel 60 plates, using an elution solvent of 80-90% acetonitrile, 10-20% water, with multiple ascents (generally 3-4 ascents). The plate is irrigated to the top for each ascent, dried by hair dryer, and placed back in the tank for the next ascent. Each ascent of a 20 cm plate takes less than one hour at room temp.  This is essentially the method we described in Meyertons, J.L., Labeda, D.P., Cote, G.L., and Lechevalier, M.P. 1988.
