You can add 6%, 12% and 18% of high mw PEG (8000-10000) in the nutrient solution to the soil at  3-4 days intervals for 12-16 days during stem elongation or heading to induce mild, moderate and severe stresses, respectively

For in vitro conditions, you could use PEG (4000) AT 0,0.5,1.0%.

For in vitro callus culture you can use 1, 2 and 3% water soluble PEG-6000  for induction of three levels of water stress.

I had not realised you are working on callus. But the need to measure the osmotic potential (OP) of the PEG is still important and to consider damage effects etc, and the refs give an idea of the OP of different PEGs (including I think 6000). Why not do a preliminary experiment with a range of OP to see what the effect are?
