In addition, my favourite tool is STRUM (https://zhanglab.ccmb.med.umich.edu/STRUM/)

user-friendly online tool which systematically evaluate effect on protein stability of all possible mutations in provided protein sequence or PDB structure. Only drawback is processing time - about 24 hours, but they send you an email, when the job is done.

Firstly you can choose gene of your area of interest, then go for its SNP analysis(dbSNP), find out positions of tolerated and damaged amino acids. then choose the one or more positions which are damaging, then use I-Mutant (version 2.0) (http://folding.uib.es/i-mutant/i-mutant2.0.html) to find out the effect of missense mutations on protein stability in very simpler way.
