Dear all, I would like to know whether there are any advanced methods of determining chromosome number in plants other than traditional microscopic studies.

Nirmala SVSG in the second number suggestion from the top iz an excellent method. Just identification of a single chromosome through well known chromosome specific DNA sequence of the particular crop species using in situ hybridization the method mentioned. But, simultaneously, you can detect three chromosomes or three arms of the chromosomes by three well known DNA sequences by multi color in situ hybridization iz more powerful. DNA x chromosome hybridization after labeling with isotope or non-isotopic coloring detecting biochemicals such as digoxigenin, biotin are generally used. Please, check next labeling for more than three arms or chromosome identification or numbering. But, special skill obtained through long work and experiences will certainly helpful for this sort of numbering.

You may want to tell us what species or plant group you are interested in determining chromosome number. Is it just number or more (e.g., chromosome morphology)?
Collect seeds and try if there is no dormancy they will germinate.Harvest the root tips afyter giving colchicine treatment to get good metaphases.Try acetocarmine or acetoorcein, If the species is flowering you collect young buds of different stages in acto-alcohol or any other suitable fixarive and squash anthers and examine.
