I've got the TEM pictures of my purified bacteriophage whose host is S. aureus. there are various shapes with tail/tailless (maybe a little tail). in order to place it in a family, I need the measurement criteria. can you introduce references, please?

In my suggestion, you may use the CsCl gradient purified bacteriophage instead of the filtered supernatant. You may also modify your negative staining process using Uranyl acetate or phosphotungstic acid for better staining. you can observe the clear morphology of phages. Hopefully, it works.
Scott Nguyen No. It is from the purified lysate of a lytic phage. do you think it's possible for helper phages to form when a strain is used for propagation of a strong lytic phage? I think when it happens, the bacteria are in full possession of the lytic virus. also, I ran a PCR test on the purified lysate for the Integrase gene and it was negative.

Md. Shahed-Al-Mahmud It was purified by the PEG/NaCl process and unfortunately, I had no access to CsCl and ultracentrifugation. Also, the negative staining and imaging were done with another person and I had no access to change this part of the process.

in other TEM images, I can see the shape of a neck and tail tube, which represents members of myoviridae family. I uploaded this image here. Can I have your opinion about this new image?
What source did you use to isolate your bacteriophage ? May be you have more than one lytic phage againts your bacteria, because it seems a phage with an icosahedral heah and long noncontractile tail in part c of first image that belonged to Siphoviridae family and in second a phage with an icosahedral head and contractile tail belonged to Mioviridiae family.
The sample you prepared for TEM is not well diluted and stained.
And did your prepare sample for TEM , contain phage alone or with its bacterial host?
