I have the infectious clone mediated with agrobacterium tumefaciens with kana and rif antibiotics in the stocks. Now i want to prepare infection clone to inoculate plants. so that can any one tell me how to proceed and what amount of i should add into YEP?. thanks
Start with 1 colony in 5ml, this will serve as your preculture. Grow 36h at 28C, then transfer 20ul in 20ml

thanks for your answer. but you mean 1 colony? i do not have a colony. if i start with plasmid you are right but i have a liquid infection clone of approximately 2 ml. with this amount i want to enlarge and inoculate the plants. so please how i will do this one? thanks in advance.

you should use antibiotics to keep the selective pressure and make sure your clone doesn't lose the plamsid
