My coworkers and I have been trying to screen S. aureus for capsule production using agar plates that have been supplemented with Congo red. My coworker has done this before during graduate school, but at work it doesn't seem to be working. I really want to get these plates up and running, because they'll be useful for some mutagenesis screening we'll be doing in the near future (yes, a PCR will be done eventually on the samples, but assuming I get a large quantity of mutants I want something a bit more economical for the initial screen). We've tried two recipes so far (which are below), and I'm going to buy new Congo Red just to be safe. But does any one have any suggestions to get them working besides that? My group is kind of tapped out on this one.

1) 40g/L Blood agar base, 0.4g/L Congo red, 10g/L glucose.

2) TSA base, 0.8g/L Congo red, 5% sucrose
