I would like to know whether one has tested the BOS method (plant material dried over CaCl2), a widely used method among plant virologists,  for sampling pieces of stem or roots infected by vascular pathogenic bacteria.

THe idea would be to sample pieces (or tranches) of stems in the field, store them in CaCl2 for one/two weeks, and then isolate the bacteria in the lab using an enrichment method (dilaceration of the inner tissues and incubation in a semiselective broth) or simply after maceration in sterile water/tris and plating.

Does anyone have an experience on that ?
