I am using an 5 day BOD test on a Hach BODtrack (not the Bodtrack 2), samples taken from river in sterile bottles straight to machine.
One sample tested at 70mg/l, one at 35mg/l and the last one was a baseline which we spiked at 350mg/l but I am getting some really strange curves?

There are many different points or non point source of pollution with the flow direction.

The initial bacteria take time to grow and more refractory COD in the water such as lignocellulose type materials can require some slow growing species for oxidation. Remember BOD5 is not the same as BOD30.
Microbiological dynamics in the real world can make the results all over the map. That is why you want both BOD5 and COD. For some purposes I like BOD30 where slow growing decomposers can have their inputs.
