I'm a university student who would like to perform a small root development study. Since it's my own project I can't afford to buy the very expensive agars from experimental suppliers.
Plant propagation agar is used for growing plants in various experiments. It is slightly opaque/cloudy. When studying root growth you need a completely transparent agar. Transparent agar is mentioned in many studies, sometimes referred to as "improved agar-plate culture system" or IPG, "improved plant-growing".
However I can't find any reference anywhere to what the composition of transparent plant agar is. Does anyone know where I can find this?
If I can find the composition, a professor has promised that I can buy the chemicals from the university at cost and a doctoral student will let me use the microbiology lab equipment.

With the few details that you provide, some questions arise. Is it necessary to use agar? If you need "to see" how the roots growth, you can use liquid media: using paper supports you can keep stand up the explants, with the basal portion in culture medium while the other part is over it. On the other side, if your purpose is to obtain just rooted microshoots, you can use other kind of supports as vermiculite, cotton, rockwhool......
yo utilizo para ver la formacion de raiz, uso agar chino (utilizado para comidas) como son explantes pequeños, con 6-7gr/L es suficiente... lo otro es que utilice gelatina, como usted dice: "solo para observar raiz".
Yo en mi caso despues de hacer estas pruebas y ya quiero hacer algo más riguroso utilizo Phytagel o gelrite (le tengo más fé al primero) el medio es transparente.
