When I focus the microscope the surrounding parts remain the same but the marked point changes with changing focus of the microscope. Can you help me to understand why this happens? What is there?
As for others to me, it appears clearly to be an air bubble. The change with focus is related to the relation between the section thickness and the bubble diameter. The darkening around the air is linked to refraction at the air / water interface. Fairly typical.
If you have any suspicions about your object you should repeat experiment with some color for better visualization of your emulsions or to label continuous phase. This marked object really looks like air bubble. I usually use the cover glass when make the photos of emulsion droplets.
