My ligand has lambda max of 258 nm and CT-DNA is 260 nm. It is currently unknown if the ligand or ligand-DNA complex possess fluorescence due to instrumentation issues. With the wavelengths being so close, is UV-vis even an option to use to determine the stoichiometric ratio between ligand and DNA? If it is determined that there is no fluorescence later, what other option would be best?
Michael, maybe do a curve of measurements(UV and Fluorescence) by adding known concentration of ligand to known concentration of your DNA? Titration! To see any abs increase and Fluorescence increase.
Why not run molar absorptivity check at different concentrations of both the standard and sample and them together at your fixed absorption maxima for both, the analyte and the standard!
