we would like to perform immune histochemistry to stain antigen-presenting cells on porcine lymph nodes. I found that SLA II DR and DQ are options, but which one would be best? Are there alternative options? Thanks!
If there is homozygosity in the model animal either DR or DQ you can chose easily. Maybe you can do single staining of DR and DQ and double incubation with both DR+DQ.
Altnernatively you could stain other antigen presenting cells with antibodies against dendritics or macrophages ...
Do you know about antibodies that can be used for more specific staining of dendritic cells or macrophages? I was actually looking into the DR and DQ antibodies as I couldn't find any more specific markers.
There is a large number of antibodies against human dendritic cells (professional APCs) that cross with animal species.
The CDs most commonly studied are: CD 204 to CD 209 for lectins, CD 283 to CD 289 for TLRs, CD 303 and CD304 for circulating DCs, then CD 467 and many others ....
Example of clones crossing between humans and pigs:
CD 207, langerin, clones 929F3 or 310F7, langerhans cells.
CD 208, DClamp, clone 1010E1, mature DCs (pulmonary DCs)
