I am working on human iPSC-derived neurons and astrocytes. As part of my project, I am attempting CRISPR-mediated knockout and HDR knock-in in these iPSCs to target specific genes. However, despite multiple attempts, I haven’t been able to achieve successful knock-in results.

I would really appreciate any advice on troubleshooting or suggestions for improving the efficiency of these CRISPR experiments. If anyone has experience with CRISPR editing in iPSCs and is willing to share protocols or insights, it would be incredibly helpful!
