It depends on the type, or in this case polarity, of secondary metabolites you are interested in extracting. A simple way to extract compounds is with maceration, which involves grinding up the plant material to a powder (let's say plant leaves to be specific), adding a solvent such as water, letting the mixture sit and agitate for some time and filtering via pressing or centrifugatoin to remove plant particles. Heat can be applied following the solvent addition. Here is an example article in which this maceration procedure was performed with Pinus radiata bark:

However, note that this non-specific technique will simultaneuoisly extract many different compounds according to their polaries in relation to the solvent polarity.

It depends of the quantitative measure and the textures of the plant. There are several ways, distilling, using saline solution with the end step as heating or boiling the saline solution until extractions are forced to appear on the top layer of the saline solution. The top layer can then be suctioned out and stored for a certain amount of time.

Aqueous extraction

10 grams of dried plant powder was left in distilled water for 6 h at slow heat. After 12 h it was filtered through what man’s No.1 filter paper and centrifuged at 5000 g for 15 min. The supernatant was collected and concentrated to make the final volume, one-fifth of the original volume. The extract was then autoclave at 121°C and 15 lbs pressure, and stored at 4°C.

Solvent extraction

10 gm. of dried plant material was extracted with 100 ml of methanol and was kept on a magnetic stirrer for 24 h. Thereafter, it was filtered and centrifuged at 5000 g for 15 min. The supernatant was collected and the solvent was evaporated to make the final volume one-fifth of the original volume. It will be stored at 4 °C in airtight bottles for further studies. The same procedure was repeated for obtaining ethyl acetate, methanol and chloroform extracts.

actually, I know all this method, but I search a new method; for example, I want to isolate only sesquiterpen and terpenoid from plant material or only phenolic compounds. I dont know there is any metod for that...

sorry for that ,if u want to isolate only sesqiterpen and terpenoid y dont u go for HPTLC PREPARATIVE after soxlet or microwave method for isolation of uer compound

I have some endemic plant materials, I suggest that they have some cytotoxic and antimicrobial compounds, however, if I do only soxlet extraction or maceration, the result will not enough importance for me...

now, I dont have a phytochemistry lab. and there is limited condition...

if I dont find anything, I will do that ;

1- xylol extraction, I will get filtered material (pelet)

2- pelet in dichloromethane extraction, I will get pelet

3-pelet in chloroform extraction

4-pelet in methanol extraction

I think that it can be better than only one solvent maceration...

it is nice to see that someone read my post. I will get this post for further hypotheses and secondary test to see the best result. I will check back from time to time for discussion purposes. Creativity and ideas work well with limited resources and finding the best possible solution without so much filtration. Filtration paper can filter so much. Rehana way would have been better for herbal medicine and not treatment of depleted soil and micros to plants sometimes considered pest.
