When we introduce probe into the system such as ss-DNA, there is a transition from random to ds-DNA(rigid) or say for example duplex or even triplex etc. Can somebody please quantify(with appropriate justification) why it is relatively difficult to quantify the effect of such perturbation on the structural configuration of DNA/SS DNA/ds-DNA etc. through structural or physical tools such as XRD etc. (Even though there is dimensional changes.)

Because i am unable to find any appropriate literature on that.

Because even though u can quantify these through other means such as electro-chemical approach. Can somebody put a light on the structural quantification of config-rational switching through various material characterization tools such as XRD. or suggest some appropriate literature on that .
