I am planning to record compound action potential (CAP) in stellate ganglion nerve trunk. The approach I will use was from previous labmate. Here was the descriptions for the electrodes used in CAP recording: Stimulation and recording electrodes were made from Teflon insulated Platinum Iridium (PtIr) wire of 0.004” with 0.0005” sheath crimped into adjustable gold connecting pins and were separated by 0.6mm.

Here is my question regarding this method:

1, Do both stimulating and recording electrodes need two wires?

2, Do I need to twist the two wires? How to build (secure) the two wires separated by 0.6 mm, if the two wire can not be twisted together?

3, Do I need to remove the insulated Teflon in the both ends of the wire for a good conductivity?

3, Will the Ag/AgCl wire be replaced by two Ptlr wires for the recording electrode? What solution should be filled into the recording electrode?

I never had a experience for CAP recordings. I would appreciate if you could give some advices and suggestions regarding the questions.

Thank you very much
