The Lipinski "rule" of five is more of a guideline than a rule. There are many drugs that violate the rule but still have excellent pharmacokinetic profiles. First of all, the molecular weight of your compound is not that big and good absorption would still be acceptable. The logP value does not make sense for intestinal absorption since it is far over the desired range.

1) How did you know that GI absorption is high?

2) I can take a look at the structure and try to make an educated guess as to why the absorption is high.

hello dr. ,,i have manufactured compound when i put it in ADME which is abbreviation of absorption, distribution, ect...found 2 violation in lipinski,also 2 violation in chose and 1 violation in mugge in m.wt =560 and mlogo=4.27 and xlogp=7.08 but TPSA still 88 not exceed 140 and GI absorption appear high and i am unable to explain why GI absorption stay high despite the presence of violation,,thank you very much@ Rafik Karaman
