I have trouble with getting proper receptor expression with paraffin section IF, would anyone will like to suggest that I should try more or move to frozen sections?
In order for the RG community to provide suggestions, we need to know a little more about your receptor(s) of interest and the technique(s) you would like to use.
Thanks for comments and I apologize for not clear enough questions. I am using heart and vessels sample and checking Sphingosine 1 phosphate receptors.
It really depends on your antibody/antigen pair. We've done lost of IHC in frozen or paraffin embedded sections to detect GPCRs (like yours) with mixed results. Some work fine in both, some work better in frozen tissues. Honestly, frozen sections are easier to handle and are less manipulated -no washing with organic solvents, for instance, and I have never seen an antibody that works with paraffin sections but not with frozen ones. The converse, however, is not true. So, if you have a choice, go for the frozen sections.
