Im using 2,4 D , Kinetin, BAP for callus induction and somatic embryo genesis of an woody plant. I"m using the above hormones above. Is those hormones are autoclavable?
An interesting related point that I did not know before (perhaps the rest of you do): some PLANT hormones may be useful in treatment of cancer in HUMANS.
We are using BAP and IBA in our propagation medium for elms. All medium ingredients were autoclaved together. We had never any problems with that method.
Generally synthetic harmones are autoclavable. 2,4-D and BAP are also. Kinetin is natural and it is heat liable. you can autocalve but u have to add more. Best option is to filter sterlize all these harmone.
I agree with u dr. Ravinder . Regarding the GA u can dissolve it in absolute alcohol, than u can add to medium after autoclaving. But keep in mind this thing must be achieved under aseptic condition. IAA is autoclavable but it can be sterilized by filtration
I would use 95% etoh, NOT absolute as it has an organic solvent added (generally Benzene) to remove/capture H2O and is therefore poisonous and may cause issues with tissue culture (remember we use only 70% etoh to sterilize). And YES, you can autoclave the synthetics, but sterile filtration removes "floaters" and is quick and easy. :)
A millipore filter (0.22 µm) can be used. Empty glass is sterilized in an autoclave before filter sterilization. prepare a required aqueous stock solution of GA3 (mg of GA3/ml of D.water). sterilize the medium by autoclave,then cooled to 50-60 °C before adding GA3 under aseptic conditions.....good luck...............regards
sir i am using NAA and GA3 for exogenous application how i can dissolve them i tried to dissolve in ethanol and then in distilled water but precipitates are formed
One can dissolve hormones in DMSO and add to stelie medium after autoclaving. Only one pre-caution_ final DMSO concentraztion must be less than 0,5 µl/ml.
But all hormines can be dissolved in DMSO at high concentration.
