I am dealing with Phosphate solubilizing bacteria. They produce different types of organic acids and secrete acid into medium. This acid can solubilize the complex phosphate compounds into orthophosphates. My question is why does they produce these organic acid and secrete out side from the cell?
Dear Pasindu, major mechanism of mineral phosphate stabilization is the action of organic acids synthesized due to physiological activities by the cell of phosphate solubilizing microorganisms. Production of organic acids results in acidification of microbial cell and its surroundings. As the microbial cell dont have the capacity to retain more than 15-20% of the organic acid produced by them, most of them are come out extracellularly. Consequently Pi may be released from a mineral phosphate by proton substitution.

In media you get production of organic acids that depends on the carbon source you supplied. eg. Bacteria mainly belonged to gram negative, possible reason because they possess glucose dehydrogenase (GDH), which allows glucose to be converted to gluconic acid in the periplasmic space. Same bacteria may produced different type and amount of organic acids depend on the C-source supplied in to the media.

Perhaps you should always think about natural environment, in soil phosphate is available in insoluble or in more complex form which is not useful for bacteria/plants, to make it useful bacteria produce/secrete organic acids to convert insoluble phosphates to more useful soluble phosphates for their metabolism or to make it available for plants. same thing is applicable to in vitro experiments.
The environment creates the need, the need creates the function and the heredity consolidates it.

I already prepared modified pikovskaya media by using iron phosphate instead of tri-calcium phosphate but the media is quite transparent and I think it is not possible to determine the zone. Is broth culture with iron phosphate will be good ?
Chandra Sekhar Paul try this: If you can not see the halozone, after incubation stain the plates with 0.1% congo red or grams iodine keep the plates for 15 mins then wash it with NaCl (optional use 1M NaCl), try to measure the zone. Perhaps you can also try iron solubilization in liquid media. I hope this technique will work.
