Hello, I want to estimate the total carbohydrates from my sample
I actually took 100mg of sample and hydrolyze by adding 5 mL HCl, after that I made up a total volume of 20 mL (Sample volume)
I took 0.2 mL from each sample and added 0.2 mL of phenol and 5 mL of sulfuric acid.
The absorbance I got ___ 1.125
from standard curve of glucose the concentration comes 0.654 __ by the formula =TREND(Conc, Abs, Sample)... that way,..
Anyone, please help if I am doing everything right... and how can I determine the TOTAL CARBS (%)... or mg/g value etc..

I would suggest you to use anthrone method for estimation of total carbohydrates. Donot use phenol here, construct standard curve of 1mg/ml of standard glucose and estimate your total sugars from your sample .
Dissolve your sample gm/ml and take known volume to estimate, then you would get total sugars
