Hi everyone,

Can someone please explain to me how to calculate the p-value of the fold change for gene expression.

I made an experiment with 3 controls and 3 treated samples. To analyze relative changes in gene expression (fold change) I used the 2-ΔΔCT Method. For the untreated cells i calculated 1. (control --> no change --> ΔΔCT equals zero and 2^0equals one)

So for control i have:

1

1

1

And for treated cells i have a fold change of:

6,32427088

3,71218144

4,5445967

I tried a T-Test in Excel but the results of th p-value are clearly wrong.

This is my first time calculating p-value of fold changes. After some research i found that sometimes they use the logFC. I tried this to but the results still can´t be correct.

I am pretty sure I make something wrong but i can´t fiqure out what it is....I would be very grateful if someone could explain it to me for beginners.

Thank you!
