I'm wondering what the most suitable non-toxic CsA and RuR concentrations are to treat neurons for six hours. The concentration should be sufficient to block mPTP opening and MCU-dependent mt-calcium entry as well. I'm using near-pure primary cortical cultures.

Rutenium Red is not cell permeable, try using RU 360, the problem is that it is very instable, and its action lasts short times, we have used it at 10 uM in intact neurons and it works, at least for shorter times. CsA is less problematic, 1 uM should work fine in neurons.
