Currently, I am studying the immune response to exosomes isolated from patients' serums. Is it sufficient to standardize the amount of exosome based on the size of the blood sample that was started? It is known that the number of exosomes is not constant. Is there another solution without having to purchase a kit that counts exosomes? protein or RNA concentration ?
From your experience, what is the sufficient volume of serum to induce an immune response to cells such as dendritic cells ?
Also have you done MTT test with exosomes ? can you share your thoughts on that please
Yes, you can measure the protein concentration in the exosome sample and add fixed amount (mgs) of proteins for each sample. People uses this alone or in combination with nanoparticle track analysis if available.
MTT depends on measuring the metabolic activity in the health living cells which should be absent in exosomes. Unless you mean you are going to use the MTT to test the effect of exsosomes on the viability of the cells?

and cytotoxicity before checking other parameters I want to study such as cytokines releasing from immune cells ( dendritic cells).
