I am planning a cell culturing project where I plan to introduce expression of one gene to a cell population (cilliary neurotrophic factor; CNTF). The original article used LZRS lentivirus as the transfection vector. How important is it to use the same vector when trying to reproduce another lab's results? I have been trying to design this plasmid, but have been having trouble finding appropriate restriction enzymes for constructing the plasmid. Is there another vector that is more popular/cheaper/easier to work with/more efficient than LZRS that I can use to introduce this gene? I am also looking to introduce a TROY mRNA RNAi plasmid into another cell population. Is there a highly efficient RNAi vector that is inexpensive that anyone recommends?
