Measurement of carbonic anhydrase in haemolysate?

We have been trying to measure this but are finding the Haemoglobin is interfering with the assay.

We removed the buffy coat, wash with 0.9% NaCl and lyse with 4 parts water. p-nitrophenyl acetate is the substrate.
