We are conducting multiplex immunohistochemical stainings on pediatric liver tumor samples. We are trying to phenotype macrophages by differential marker expression. One of the macrophage markers we are interested in stains LSECs as well as macrophages. Since macrophages are predominantly located in liver sinusoids, adjacent to LSECs, we need an antibody that specifically stains LSECs, but not Kuppfer cells / macrophages. This way we can differentiate between macrophages and LSECs and confirm that the markers we are interested in are indeed present in macrophages and not just in the adjacent cells.

We have had little luck with finding a working antibody. The ones we have tried are anti-L-SIGN ( or anti-CLEC4M) and anti-AOC3. We have tried multiple antibodies from different companies targeting these markers, since finding a LSEC-specific marker in general has been challenging.

Does anyone in the ResearchGate community have any suggestions in for an LSEC-specific marker?
