Hello, several reasons can cause problem.

Check injection needle, injection valve, leak in system, etc.

More info for more exact answer, do you have normal signal for reference standard or any other component ?

Thank you for answering Mr Karen. I dont think is injection need or injection valve. There is no leak because it has been checked with other column and analytes. I am trying to run using standard but could not get appropriate signal at first. When I changed to Agilent from Shimadzu, I had some result but it is not dose responsive and could not have separate peaks for standard and internal standard. Now the agilent unit is broke down and have to shift back to shimadzu.
