(Difco, USA) for Phytophthora infestans sensu lato, and added different antibiotics, and these were incubated at 16°C with a photoperiod of 12 hours of light and 12 hours of darkness.
It’ s so easy to transfer inoculum (Mycelia) from tomato fruit to tuber disc of potato (previously sterilised with hypochlorite of sodium at 0.2% than washing with distilled water, incubation at 20°C for 4 days .
Concerning leaves, we can placed it in moisture roomat 20°C, for sporulation.
After confirmation of the presence of no septate mycelia and sporangia of Phytophthora after isolation and microscopic observation . We preleved developped mycelia into petri dishes containing Pea- agar medium (125 g of pea boiling in 1000ml of water + 15 to 20 g of agr), we can added antibiotic to eliminate bacteria contaminants. The incubation is in the same precited conditions. Good continuation.
